My only thing with huge leagues is don't make the rosters big as well. Keep it standard (qb/2 or 3 wr/2 rb/te and not a bunch of flex spots, maybe one flex that can be anything), because I've been in 20 person leagues with big rosters before, and when someone goes down you're looking at the 4th RB for the Pats or some ridiculously little-used player to fill your 2nd flex spot. I'm not opposed to a big league, though. It's more fun and weeks are influenced a ton by those low-round picks who blow up (think Laurent Robinson from last year), moreso than smaller leagues where every starter is a probable stud player.
I'm a huge proponent for the least amount of positions as possible. I feel the more you have, especially flex, leaves more room for luck and less room for skill especially in deeper leagues. I usually prefer 1 QB 3 WR 2 RB 1 TE 1 K 1 DEF
As for the TE discussion, I think there are 20 serviceable TE's out there. I would however propose some sort of cap on how many TE's you are allowed on your roster to prevent hoarding. If someone was to some how draft Gronkowski in the 1st, Graham in the 2nd, and Finley in the 3rd. They would have an incredible amount of leverage and still draft a competitive team.
I'd also like to see a bench of 8 or more but that's just me. I like to be able to take a chance on a young player or two that might break out during the year. Also, between injuries and bye weeks it can get tough with a thin bench.

Basically what I'm getting at is, if some one were to draft 3 premier TE's, in a 20 team league, it would almost be to the point where a good part of the league would be in a potion that the position would be a waste. Combined with byes, I think it has the potential to get ugly. This isn't same with other positions. Even the 30th ranked QB in the league puts up numbers weekly, same can't be said for maybe even the 20th ranked TE. Obviously it's a huge hypothetical but it's just a suggestion. I personally have never played in a deep league so I am just trying to think of ways to keep things interesting and fun.

That way, if you don't get a good TE, it doesn't really matter. Just trying to think of ways to prevent the TE spot from being an issue. Then a K, D, and a 10-man bench?
